Some great shouts so far. I'd also add:
    LA Story
    Ted
    The Hangover
    Talladega Nights
    I Went Down
    The Nutty Professor

Kung Fu Hustle. It's an affectionate parody of all those Kung Fu film that have a big of comedy in them.

Has also been described as Jackie Chan and Buster Keaton meet Quentin Tarantino and Bugs Bunny.

I agree it's a great film, but I don't really think of it as a comedy--more a coming-of-age story with some funny moments.

Hot Fuzz , Just so much foreshadowing. It's worth watching again for the greater good.
The Rocky Horror Picture Show - apparently some people like this
Monsters, Inc. - don't forget the out takes at the end
    Delicatessen
    Sleeper
    The Wrong Trousers

My taste changes depending on mood but all I can think of for this thread is In Bruges. It's obviously quite dark for a comedy but some parts are genuinely hilarious. I dont normally like Colin Farnell but his delivery in this is excellent.

    Totally forgot about The Room.

The fact that it's supposed to be serious makes it even funnier.

Hard to make a definitive list.

Blazing Saddles, Airplane, Groundhog Day, Dumb and Dumber all obvious ones.

Not sure if these ones have been mentioned-
    The Odd Couple
    the man with 2 brains
    Clerks (presume mentioned)

A fish called wanda wasn't been mentioned yet and How to get ahead in advertising is another decent comedy featuring Richard e grant.

I thought Borat and Bruno were hilarious, probably not to a lot of peoples' tastes though.
