How far apart should the sub-structure floor bearers be placed in a timber frame shed measuring 10 ft x 6 ft? I'm told they should be 400 - 500mm apart.

Span tables will tell you what you need to know,
Also depends on exactly whats going on the floor, and the floor material you use.

Normal shed is probably going to be ok with 400mm centres.

But consider the other points as well first.
