I've a 2010 iMac with three 2GB sticks of RAM installed.

Can I purchase a 4GB stick of Ram and install it in the final slot, or do all the slots have to have the same amount of RAM?

I doubt it, only high end Macs like the Mac Pro are sensitive to this, in that they won't run at all, or they ignore the RAM in some slots.

You should be able to run varying sizes of RAM in an iMac, but best to download this app http://mactracker.ca and check your specific machine...

It will be fine. This was only an issue with much older Macs.
