How do you know? Is it sounding rough or did a mechanic tell you?
Usually coil pack is the culprit.

Take spark plugs out and verify they spark.

As biko said, start with spark plugs and work back.

When you pull the spark plugs out (one by one) check for oil inside.

I had to read that one twice, you mean off on a cylinder I presume?

As said above, remove the plugs and, normally, it will be obvious which cylinder is misfiring as one plug will have different coloured deposits to the rest.

Then, if you identify the bad cylinder, you need to move on to fuel, spark, compression to see which is missing.
