It depends.

If your an old man, a wink generally follows some sagely piece of advice.

From a younger man, a wink towards a fellow male indicates that he's in the know about some confidential information. But towards a girl it is used as a greeting.

When a woman winks at a lad though, it generally means she is looking for some sexy time (my knowledge of this is based solely on the porn industry).
