Get a road bike if you don't already have one and ride the wheels off it between now and the hundred. Make sure to work in a shedload of hills. Stop eating any garbage and get padded shorts and a cycling jersey if you don't already have them. That's about it.

If you're moderately fit you can probably do 100km on the flat without training provided you take it handy. Hills will mess with you completely if you don't train for them. That's my experience.
Assuming you've a month or so to practice, start off with just getting a few long rides in, 30-40km and work up fairly quickly from there. As with any exercise, don't do back-to-back long rides, leave a gap of a day or so. Rest is important. Running/swimming/skiing/couch-surfing all use different muscle groups so don't assume that if you go for a bit of a run you'll be fine 'cos it's not quite the same thing.

Find out how long the climbs are and find a hill near you that you can train on, it doesn't need to be as long as you can speed down and tackle it again to simulate a longer climb (kinda). Just getting some experience of tackling steep, long hills will help, even if it's just so you don't get psyched out on the day.

The biggest problem on a long ride is the pain in your ass. Definitely padded shorts and some cream to help stop chaffing. Getting time in the saddle is really the only thing that works. No matter how you cut it you will have to get used to the saddle or suffer on the day. Don't get some big-ass padded saddle, it'll do more harm than good.

Cycling in a group is a job in itself and I've never been very good at it. The great and good suggest joining a club and learning there. Sounds like good advice :)
Not knowing how to ride in a group can lead to accidents, particularly when you get tired so try to get some training somewhere or at least read posts here and elsewhere on the things you need to know.

If you try to ride with a group that has done this sort of thing before you will get dropped pretty quickly, certainly at the first real hill. Make sure you don't end up on your own/getting lost/broken down. I don't know who is organising the event but me sure they are aware that its your first time.
Learn how to fix a puncture or change a tube if you don't know already. If you haven't fixed one in years try a few dummy runs, just in case.

Beyond all that, as others have said, get a racing/touring bike and train on the bike. Best of luck!
