Potential to fail trials

Given the very strict guidlines for what can pass and fail in Pharma, I am curious about historical medicines.
Medicines that were around before big trials were the norm.

I am curious based around the Alternative proponents being asked (rightly so) for evidence that treatment works or is not dangerous.

Can anyone think of everyday medicines that would struggle to pass todays trials?
Conversly, any medicines that failed, based on seemingly innocent enough side effects that are present in the older medicines

Maybe not name branded medicines in conjecture like this?

There is no way aspirin would get through!

Don't most of those drugs, including aspirin, feature in standard-of-care controlled trials?

The ones that I was specifically thinking of were ethanol, aspirin, caffeinne and nicotine.

Atomic, Never heard that there was a batch of trials done on everyday items.
Would love to see some of the work on that, will have to google it later

I would have thought aspirin would probably fail in trials for OTC analgesia - too much GI bleeds etc. But probably not as an anti-platelet drug.
