Internet connection existence with more than one proxy

At work,we have got ISA 2004 (FQDN: proxy.rami.ae,,,,,fake name ) which has two interfaces,

NICA 192.168.1.250 connected to core switch 4500

    NICB for DMZ area ,,192.168.101.294

From DMZ through PIX to outside world.

I believe that all internal PCs (workstations) go to internet through ISA, the reason (Is it right conclusion ? ) I am saying that because we have to configure proxy within Internet Explorer -- > Internet Option à Connection à LAN setting

    proxy,,,,,,,,,,,,,,,,,,port 8080

Some times when there is lack in the internet connection , I changed the LAN Setting to :

    proxy1.telecomm.net.jp (fake one),,,,,,,,,port 8080

And some times If I chose the option :,,,,,,,,,,,,is this some thing related to a browser cache ?

“ Automatically detect settings “,,,,it worked as well

How can we troubleshoot (step-by-step) that ? (i.e how can we force a PCs to use the right proxy ?)

I'm not 100% sure what your question is.

You could force the proxy settings via group policy.

Shouldn't this all be isolated by routing rules.

Not sure i understand the question though.

How to get IE to choose a proxy or DIRECT or blackhole

IE has a 30 minute cache for proxy server or reboot.

IE on autoconfig will pickup proxy setting from http://wpad/wpad.dat OR DNS OR DHCP (note you have to setup option 252 AND a trailing space)
which it uses depends on which version of windows

The question is, how can I force all PCs to use the proper proxy which is : proxy.rami.ae ?

You meant something similar to this , will do the job :

You can force them by routing if you want , basically whatever is on the way to the internet will only allow webtraffic out thats been through the proxy . Mind you don't setup a proxy loop by accident though.
