No, you don't have to empty your account first .. you just have to make a lodgement, doesnt matter what you have in your account at the time you make the lodgement.

Usually one of the T&C's of a re-load bonus says that you cannot avail of the re-load bonus if you have withdrawn money from the site in the period after the reload bonus started and before you actually deposit money onto the site.

The best thing to do is, put money in and then withdraw it later. Although again this might nullify the bonus. Check the T&C's of the offer.
