Is it showing up in Start->Control Panel->Administrative Tools->Computer Management->Storage->Disk Management?

    Sillyaspie: You're just unfunny. hush.

    shanethemofo:
Check if it shows up: Right-click on My Computer -> Manage -> Disk Management
Assign letter / Format as required (Format = data loss. Ahem.)

I had the same problem up until recently. I had loads of networked drives and my pen-drive couldn't be found anywhere, so I disconnected one of the drives and refreshed My Computer, tadaa, there it was.

Restart the computer if you haven't already, might take effect after that.

as neuromancer said, you probably have mapped drives mapped to drive letters at the beginning of the alphabet (E,F, etc). Disconnect these, and map them through letters like X,Y,Z. You should then see your USB drive mapped to E,F, etc after a reboot.
