If a club have 40 players should they put in 3 teams for 7 a side at u10 level or 4 teams ?

I'd say if you have the coaches then go for 4. More game time for everyone.

As far as i know, the maximum squad size for 7v7 is 12?

If you have the coaches, go for 4 teams to maximize game time but that would depend on other variables, for example, if your club streams players on development level at that age (most clubs do) etc.
