I plan to travel to Vancouver September and my flight stops in New York at 2pm and my flight to Vancouver isn't until 12:40am. Is it possible to leave the airport to do a quick tour of the city ?

Yes, the US authorities do not quarantine transit passengers so between flights you are free to leave the airport. Which in turn means that whether you plan to leave the airport or not, you will need to have your ESTA sorted. You should specify 'in transit' as your destination in the US.
