I see there's a number of operators plying this route: GoBus, Citylink, Bus Eireann. Which offers the most comfortable service? In terms of seat-comfort, legroom, AC, wifi, etc...

    The train, obviously.

Absolutely not!! The train is more expensive, stops (the direct buses don't). often takes longer, and keeps letting people on even if there are no more seats.

I think there's very little between Citylink and GoBus, depending on which departure time suits better. I happily switch between the two.

GOBE is gobus and bus eireann.

Gobus seem to have good decent buses with toilets.

OP asked for comfort, not price, speed or amount of stops! ;) That said, OP also asked for bus service...

Personally I prefer Gobus but there's really very little between them and Citylink. Bus Éireann usually don't even get a look in...

edit: though to be fair I've not been on any of their new buses, they might well have improved!

GoBus, then Citylink Express/EirEagle, then Citylink Commuter.
Seats and a/c are equivalent for the first two.

    Avoid Bus Eireann.

Though, are you referring to traveling the full city-city distance or part way?

The Bus Eireann buses on the 20, x20 routes routes are a mixed bag, often contracted buses.

They take the old road between Galway and Athlone, with all the shakes and turns for most or all the way. Rarely have toilets if that matters. Can be crowded between Galway and Loughrea.
Often half an hour late getting out from Ayre Square to GMIT due to Galway traffic in the evenings.

The citylink commuter buses on the same route haven't been affected to the same extreme. Citylink was once an hour late during the january snow. Bus Eireann x20 was an hour late last week, during mid terms. Not even one of the windy days. And it wasn't a 3rd party bus.

The wifi can be pretty poor in the Roscommon, east Galway section, not really a surprise as so are the national radio FM stations.
It'd be nice if they'd increase the size of that tiny little "I agree" checkbox on the hotspot page for the wifi; even fully zoomed, it can be a nuisance to activate.

Not that I'd want to rant or anything.

I notice that gobus have a fleet of new 151 volvos

And city link bought a lot of gobus's old coaches (12 regs) and are now in eireagle colours.

There's a long thread about the difference over in the Galway City forum - http://www.boards.ie/vbulletin/showthread.php?p=89165403

Rough answer is CL express and GB are much the same.
