I intend to plug my PC into my TV via HDMI (connecting to an R9-270X video card).

Will audio be sent through the video card as well, or do I need separate audio cables plugged into my onboard line-out?

HDMI will carry the audio signal as well, if you were using VGA or DVI you would need seperate audio cables

Is that called "DDMA Audio" ? .. Do I need to select the video card as the audio-output device, or is it pretty much plug n play?

It should be plug n play, but just to be sure go to sound in control panel, and set HDMI as the default audio device
