I have the LE40C530 and it plays .mkvs perfectly. Transfer to USB stick, insert it into tv and media-player immediately pops up. Couldnt be easier and have never had problems with it.

I've had a mixed experience... one worked, the other didn't support the audio format. I've downloaded the latest firmware, but the USB doesn't pick up on it and so I can't update. Any ideas why?

MKV is a file container ONLY !
The Samsung may not support its audio codec e.g. FLAC or ogg , the video stream may be of a to high bit rate to play back. Easiest thing is to reprocess it using some sort of software.

As for the firm-ware try reformatting the USB in fat32 / fat16 as some TV's can be touchy in what they "see"
