Wife is setting up new business and will register for tax purpose.
Is this something she should do immediately or could she set up and start trading first.

Trade away, but you might not be able to charge or reclaim the VAT until you have a VAT number.

Assuming its an ordinary business and licences aren't needed for the sale of alcohol, drugs, rare species, firearms, nuclear weapons, etc.

Get her to consult an accountant. She can ask him whatever questions she wants and will probably receive some good advice re. setting up business.
