Have a SKY+ box in one room and an ordinary SKY box in another both connected with phone lines.
I now need to swap them over.
Can I just unplug them, swap them and reconnect in their new rooms or is there something I need to do/check first etc?

If your keeping the same cards in the box yu can just swap no problem as long as you have 2 coaxial cables going from the dish to where your moving the Sky+ box to.

Not moving the cards but I've just checked the coax cables and there's only one going into the ordinary SKY box (2 in the SKY+ box room) and no other coax cable where the ordinary SKY box is?

Well then you won't be able to record two shows at the one time or look at one show and record another unless you run a cable or move the cable.
